[Abstract]

Gonadotropin releasing hormone (GnRH) is a 10 peptide secreted by nerve cells of hypothalamic arcuate nucleus, which combined with GnRH receptor (GnRHR) in the anterior pituitary to stimulate pituitary gonadotropin cells to secrete gonadotropic hormone (GnH). Apart from playing a role in the passage of hypothalamic ? pituitary ? gonadal axis (HPG), GnRH was found in many organizations out of the brain,which plays a role through autocrine / Paracrine Mechanism as an important media combined with other biological factors together to form a complex signal transduction network.GnRH-based autocrine /Paracrine system is also exist in a variety of human tumors.GnRH was found to express in ovarian carcinoma, endometrial carcinoma, whether are there GnRH expression in cervical cancer as one of gynecologic malignancies? Currently not very clear.Many studies on GnRH were about the adenocarcinoma cells at home and abroad, there is a few of studies on squamous cell carcinoma. This study is to detect GnRH expression in cervical squamous cell carcinoma by immunohistochemistry and in situ hybridization method to explore their clinical significance.In this study, we collected 15 cases of cervical squamous cell carcinoma and 10 cases of chronic cervicitis specimens.we detect the expression of GnRH on the level of protein by the method of immunohistochemistry and the expression of GnRH mRNA on the level of nucleic acid by the method of in situ hybridization,and also, compare the differences between the relative content of chronic cervicitis and cervical squamous cell carcinoma through image analysis methods.The expression of GnRH and GnRH mRNA in cervical epithelial tissue of chronic cervicitis were all negative.The expression of GnRH in cancer cells were positive, positive substances located in the cytoplasm of cells, nuclei with light colored or not, the positive rate was 80% (12/15),which has significant difference compared with that of chronic cervicitis (P 0.05).Image analysis showed that the differences of unit relative content of GnRH and GnRHmRNA in cancer and chronic cervicitis organization are significant (P 0.05).The inconsistencies of positive rate in the level of transcription and translation may be caused for the reason:from the positive GnRH mRNA expression of squamous cell carcinoma, we can see the cancer cells can express GnRH by autocrine. GnRH expression was higher than that of GnRH mRNA shows that apart from the autocrine mechanism the tumor may affect the expression of GnRH by paracrine,and other channels.These results indicate that cervical cancer also has its own synthesis of the protein of GnRH, which suggest that GnRH may increase on the surface of squamous cell of cervical cancer or may be involved in the proliferation of cervical squamous cell carcinoma. GnRH may be involved the regulation of cervical squamous cell carcinoma, it needs to be further confirmed whether it will participate in the regulation of the development of cervical cancer. This provides a theoretical basis for GnRH analogues as a medical treatment of cervical cancer. It is not yet clear about the anti-tumor mechanism of GnRH in cervical squamous cell carcinoma, may have a direct inhibitory effect on tumor cell proliferation and promote differentiation, may also induce apoptosis of tumor cells. Cervical cancer treatment increasingly advocate neoadjuvant chemotherapy, GnRHa could prevent gonadal damage caused by chemotherapy. We could choose the anti-cancer target through the use of the expression of GnRH in cancer cells, which is one direction of the treatments of cancer in the future.
